DiO/DPA membrane potential detection kit

The DiO/DPA system detects cytoplasmic membrane potential changes using the principle of fluorescence resonance energy transfer (FRET), where DiO is a "stationary" FRET donor while DPA acts as a potentiometric FRET acceptor. The DiO/DPA system has been reported to produce a fluorescence signal change of >56% in HEK-293 cells and >25% in neuronal cultures and brain slices per 100 mV membrane potential change. The DiO/DPA membrane potential detection kit includes a vial of the green fluorescent membrane dye DiOC16(3) in DMSO (1 mL at 2 mM) and a vial of the anionic membrane quencher dye DPA (dipicrylamine) in DMSO (1 mL at 20 mM).

Descrition & Target
DiOC16(3) is a lipophilic cyanine dye with two C16 hydrocarbon chains. It can be optimally excited by the 488 nm argon laser line with green fluorescence emission (&lamda;Ex/&lamba;Em = 484/501 nm). The dye can be used to stain cytoplasmic membranes. Recently, DiOC16 has been combined with DPA to detect cell membrane potential change with high sensitivity. DiOC<sub>2</sub>(3) (3,3'-Diethyloxacarbocyanine, iodide) has been used for measuring membrane potentials in bacteria. The green fluorescent dye forms red fluorescent aggregates with increasing membrane potential. The spectral shifting phenomenon has been exploited to measure membrane potential ratiometrically.
